OS X Yosemite places a strong importance on the visuals of the user-interface, and has followed somewhat in the footsteps of iOS 7 by stripping away a lot of the complexities that has bloated previous versions. In addition to bundling in four new wonderful wallpapers with the latest OS X release, Apple has also instructed designers and engineers to tweak a number of important launch icons throughout the system. In addition to those amendments, the latest Developer Preview of OS X Yosemite also introduces four new stunning wallpapers that not only perfectly show off the beauty of the Yosemite National Park, but also promise to introduce visual delight to our desktops. Named after the famous National Park in California, the OS promises to introduce a number of powerful new features, an overhauled, modern looking user-interface and a vast array of additional improvements that should satisfy even the most difficult to please desktop user. MacOS Catalina brought big changes to the Mac, including the ability to run iPad apps natively, opening the platform up to a much larger number of developers than ever before.Apple released the sixth Developer Preview of its latest desktop OS just yesterday. Users could even have macOS slowly fade between the two background images over the course of the day. No more mountains! Mojave brought a new system-wide Dark Mode, and the OS shipped with two versions of its default wallpaper to match. Named after a breathtaking spot in Yosemite National Park, El Capitan was a clean-up year after 10.10. The wallpaper ushered in a new era based on … well … mountains. Yosemite brought another UI refresh to the Mac, making things flatter and more modern. High molecular weight proteins are notoriously difficult to transfer. #WESTERN BLOT TROUBLESHOOTING HOW TO#If you are pretty sure that you haven’t made a colossal “silly” mistake, then the next step is to look carefully at your protein to determine how to optimize transfer. However, if you are working with small proteins, you have to be mindful that nitrocellulose comes in 2 different pore sizes: the 0.45µm pore is used for proteins greater than 20kDa, while the 0.2µm pore size should be used for smaller proteins. Did you use the right membrane? Most of the time you can use either nitrocellulose or PVDF membranes.Did you use the correct buffer for transfer? It can be easy to grab the wrong buffer – and let me tell you, your proteins aren’t sticking to your membrane without the right buffer in place. ![]() Did you reverse the polarity? Once you have checked the membrane, you can easily rinse the dye off the blot and proceed with the Western. In this case, you can stain the membrane with a reversible protein stain such as AdvanStain Ponceau. However, sometimes the markers can transfer more easily than the average protein, or you might want to check transfer across the entire membrane. Seeing these marker bands on your membrane is a good sign that your transfer worked. Many labs use prestained molecular weight markers that transfer to the membrane. To determine whether it is a transfer issue, you need to check your transfer efficiency. First things first – make sure you have a transfer issueĪny number of things can go wrong during the Western blot procedure that will result in little to no signal. Unfortunately, this problem is often only realized after performing the entire Western blot procedure and ending up with no visible signal or only the faintest hint of bands after agonizingly long exposure times.įortunately, if you are certain the problem is due to inefficient transfer, there are several parameters you can tweak to “encourage” your proteins to migrate and stick to your membrane. One of the most frequent problems when transferring proteins to a membrane is inefficient transfer of protein. One might say you should start at the beginning, so why don’t we start with troubleshooting Western blot transfers? Transfer conditions, blocking buffers, primary and secondary antibody titrations – there are so many things that can be adjusted! Troubleshooting and optimizing Western blots can eat up a lot of your time. ![]() ![]() The same can happen to YOUR ships however, and you can use abilities that restore morale, repair damaged parts, reduce crew losses from boarding actions, reveal stealthed enemies, and more. ![]() Any of the above can damage individual components of an enemy ship, demoralize and kill off enemy crews, potentially causing an enemy ship to mutiny and flee the battlefield, or leaving it a drifting derelict rather then destroying it outright. You can also perform things like ramming attacks or boarding actions. In addition, because of the two-dimensional map design, you can protect weaker or damaged ships in your fleet by simply placing tougher ones in the line of incoming fire. However, as noted in other reviews, the AI is also not very intelligent, and is easily exploited in one battle after another by simply parking your fleet within a gas cloud or asteroid field, which cloaks them until revealed by enemy frigates, fighter squadrons or other means, while you safely fire back at them. The AI always knows where you are, and will generally make a beeline for your fleet, though they cannot actually FIRE at your ships if they are stealthed for any reason. ![]() You can slow down, speed up or pause the action as necessary, allowing you to more easily micromanage the abilities of your vessels, such as torpedo launchers and squadrons of strike craft, and skip the long wait while you close distance with your enemy. ![]() Story misions may have varying additional objectives and any level can have environmental hazards. You can increase your fleet cap by leveling up your overall "Renown" level by winning random battles and completing story missions. You control oddly small fleets of warships across 2D-maps against opposing fleets, both of whom are restricted by arbitrary point values. The actual gameplay in BG:A2 is much the same as the first. Furthermore, they can launch random invasions from outside the map almost anywhere, even if they have no fleets on the map, provided their threat level is high enough and you haven't knocked them out of the campaign in certain story missions. If you can't move any more of your fleets, you'll have to end your turn- and then each enemy faction gets to do the same thing. Each of your fleets has a limited pool of movement points to move between star systems, and if they attack an enemy system and win the battle, their remaining points are used up anyway. This can be resource generation to spend on new ships and planet upgrades, shipyards to build and repair ships, sources of troops to reinforce ships that have suffered crew losses, passive buffs for ships, and more. Each star system has one or more planets with various local features that can help or hinder your war effort, depending on who controls them and how developed that planet is. Any time you attempt to take an occupied system or enemies attempt to invade one of yours while your fleets are there, you'll have to fight it out in either a "Cruiser Clash" deathmatch or a control-point gametype, and you can change which one you play at any time from the settings. The campaign consists of a territorial conquest metagame with various star systems in the vicinity of the Eye of Terror, which for those unfamiliar is an area of space where the laws of reality is basically out to lunch. There are now Imperial, Necron and Tyranid campaigns, with a Chaos campaign available as DLC. New to this game are dedicated Mechanicus fleets, Craftworld Eldar, Dark Eldar, merchant Tau, Necrons and Tyranids. Far more factions are available for play: We again have the Imperial Navy, Eldar Corsairs (no, I will not call them "Aeldari", thank you) Orks, Chaos, Space Marines and Protector Tau. Jerry played through Gibson SGs and then switched to Alembic guitars. These amps were super-clean and not to my liking. He played through Fender amps all during the time I saw them. Jerry’s guitar tone was always thin and mealy so it never got me. The Allman Bros Band truly was, and then they could meander and jam, but the guitars were heavy and muscular because Duane Allman and Dickey Betts played Les Paul guitars and Marshall amplifiers. The Dead may have thought of themselves as a blues-based band but in the strictest sense, they were not that. During the jams that connected the songs, this incredible rhythm section just blew me away and, on acid, was truly mesmerizing What got me hooked on the band was the interesting (but enigmatic) songwriting style of Robert Hunter, the polyrhythmic double drumming of Mickey Hart and Bill Kreutzmann, and Phil Lesh’s bass playing. It wasn’t their singing (barely passable until Workingman’s Dead) or the guitar work (Bob Weir was straight rhythm, although with a distinctive style and Jerry Garcia…well, I never liked his guitar tone, and his lead work, while interesting, was way too repetitive for me). Who didn’t love Workingman’s Dead or American Beauty? I totally got the debut album simply called The Grateful Dead (1967), followed by Anthem of the Sun, Aoxomoxoa and Live/Dead. This is a tough one, as I have seen the original Grateful Dead more times than any other band (except my own!).Ģ7 times between 19 actually. Keith was one of my favorite guitar players but he just can’t do it. ![]() This of course would make them a “cover band” but really, the guitar playing is so lousy, why would anyone want to hear some of the greatest rock songs played in keys that were not meant to be played in because arthritis prevents the correct playing of the songs. ![]() The Stones now have a better rhythm section than they have had in years, with great replacements for Charlie Watts and Bill Wyman (Darryl Jones on bass and Steve Jordan on drums) and if Mick could do it, he should just finally put Keith and Woody out to pasture and bring in guys who can actually play guitar. In the case of the Stones, who were once one of the greatest live shows on earth, they are just plain pathetic now. ![]() The Rolling Stones, the Grateful Dead, and Bob Dylan were tougher in that I have loved these artists at different times and my issues with them, over time, have to do with what I believe is an artist’s responsibility to their fans. His voice drives me crazy and I walked out twice – at two different shows – while watching him with the E Street Band, as the show sound was awful and his stage raps were just plain boring, so that was easy. I have written critically about the Stones, Springsteen, Dylan, and the Dead in the past.
